表面等离子体共振技术及其在蛋白质活性浓度绝对定量中的应用

摘要表面等离子体共振(SPR)是一种由隐失波引发金属表面疏密电子振荡的物理学现象,通过共振角的分析可以了解分子间相互作用的情况。基于SPR的无校准浓度分析技术,通过在传质限制条件下测定蛋白质与其受体或抗体的扩散结合速率,即可在不需要标准品的情况下直接测定出蛋白质的活性浓度,是一种蛋白质活性浓度的绝对测量技术。通过综述表面等离子体共振技术绝对定量蛋白质活性浓度的原理及其在各领域的应用,探讨了该技术的优势以及目前存在的问题,展望了该技术未来发展方向及其在蛋白质计量中的主要应用。

Abstract：Surface plasmon resonance (SPR) is a physical phenomenon observed when sparse and dense electron oscillations occur on a metal surface upon interaction with an incident electromagnetic wave.The analysis of the resonance angle during SPR provides valuable insights into intermolecular interactions.SPR-based calibration free concentration analysis (CFCA) enables the absolute measurement of protein activity concentration by quantifying the diffusion binding rate between a protein and its receptor or antibody under controlled mass-transfer-restricted conditions, eliminating the requirement for reference material, which is a technique for the absolute measurement of protein activity concentration. By reviewing the principle of SPR technology for the absolute quantification of protein activity concentration and its applications in various fields, the advantages and current issues of this technique are discussed, the future development direction of this technology and its main applications in protein quantification are also prospected.
