Research on the Metrology Traceability Technology of Surface Plasmon Resonance Biomolecular Interaction Analysis
According to the core principle of surface plasma resonance (SPR) biomolecular interaction analysis, a metrological traceability method was proposed, which takes the refractive index as the theoretical basis and combines sucrose reference materials. Firstly, sucrose standard solutions were prepared and quantified using a refractometer. Then, the refractive index values were traceable to the Abbe refractometer standard block, thereby establishing the calibration basis for the refractive index, realizing repeatability detection of the refractive index, linear calibration within the range of refractive index standard values from 1.330 to 1.400, and analysis of the analyzer’s relative baseline noise and relative baseline drift. Finally, using the purity reference materials of sucrose with gradient dilution, the calibration of the detection limit was completed. For the first time, a comprehensive, scientific, and effective metrological traceability method for SPR biomolecular interaction analysis has been established, which can be applied to the metrological calibration of related instruments in the fields of basic life science research and biopharmaceutical development.
