中国仓鼠卵巢细胞宿主DNA残留检测标准物质
Chinese Hamster Ovary Cell Genome DNA Residue Reference Material for Testing
为解决生物制药产品中外源性宿主细胞DNA残留检测的计量溯源难题,大批量培养中国仓鼠卵巢(Chinese hamster ovary, CHO)细胞,研究柱层析、乙醇沉淀法、超滤法等多种提取方法,提取得到高纯度的CHO细胞基因组DNA,选择充分研究和优化的数字PCR方法作为基因组DNA的定量方法。结果表明,相比传统的UV检测,基于数字PCR的方法能有效解决基质干扰问题,结果一致性显著提升。标准物质的均匀性和稳定性通过检验,利用10家单位的5种不同的数字PCR平台给标准物质定值,最终的拷贝数浓度定值结果为(1.06±0.12)×104 copies/μL。该标准物质可促进DNA残留量检测试剂盒的开发,促进各生物制品公司DNA残留量检测结果的互认统一,实现检测结果的量值溯源。
To solve the traceability problem of the exogenous DNA residue determination method (qPCR method) in Chinese Pharmacopoeia (2020 edition), a large-scale cultivation of Chinese hamster ovary (CHO) cells was conducted. Various techniques such as column chromatography, ethanol precipitation, and ultrafiltration were employed to extract high-purity CHO genomic DNA. Digital PCR, which has been extensively studied and optimized, was chosen as the quantitative method for genomic DNA. The results showed that the digital PCR-based method effectively overcame matrix interference issues and demonstrated better consistency than traditional UV detection. The uniformity and stability of the standard substance were verified, and the copy number concentration was determined using 5 different digital PCR platforms from 10 units, resulting in a final value of (1.06±0.12)×104 copies/μL. This standard substance can facilitate the development of DNA residual testing kits and promote mutual recognition and standardization of DNA residual testing results among biopharmaceutical companies, ensuring traceability of the testing results.
